All pET vectors are based on the promoter of gene 10 of phage T7 (pŘ10) (89). Some include the rbs (S10) of this gene and those shown include the transcription terminator TŘ downstream of gene 10. In the simple translation vector the NdeI site permits fusion of a coding sequence to the rbs of gene 10, while insertion at the BamHI site generates a fusion protein. More recent vectors include a series of targets for alternative restriction enzymes – a multiple cloning site (mcs).
